Preparation Method Typical Preparation of solutions for Peroxidase Detection : Dissolve 12 mg of DAB in 1.0 mL 50 mM Tris buffer, pH 7.3 (or inject 8.3 mL of buffer into serum vials if using #150826). Working size aliquots may be stored for 6 months at -20 °C. The stock solution is diluted 1:20 with 50 mM Tris buffer pH 7.3 just before developing the immunoblot. To 10 mL add 10 uL 30% hydrogen peroxide. Add 10 mL of this substrate solution per 15 x 15 cm2 membrane. Develop the blot at room temperature with agitation until the bands are suitably dark. This typically requires 1-5 min. Stop the reaction by rinsing away the substrate with PBS.
